Reconstitution and Cold Storage: Lab Best Practices for Research Peptides

A high-purity peptide is only as good as the handling it gets after it arrives. Poor storage and careless reconstitution degrade material and quietly ruin reproducibility. These are general laboratory best practices for research peptides, for research use only.

Storing lyophilized powder

Freeze-dried peptide is the most stable form, but it still needs care:

  • Keep it cold. Store lyophilized powder frozen, commonly at -20 C for the working term and colder for long-term storage.
  • Keep it dry. Moisture is the enemy. Protect from humidity and keep vials sealed.
  • Warm before opening. Let a sealed vial reach room temperature before breaking the seal, so condensation does not pull moisture into the powder.
  • Protect from light for compounds that are light sensitive.

Choosing a reconstitution solvent

The right solvent depends on the peptide and your research design. Bacteriostatic or sterile water is common for many research peptides, while some require a small amount of a co-solvent to dissolve fully. Add solvent slowly down the side of the vial rather than directly onto the powder, and let the peptide dissolve without aggressive shaking, which can stress the molecule. Never force a peptide into a solvent it does not dissolve cleanly in.

Aliquoting: the highest-value habit

Once reconstituted, a peptide solution has a limited working life, and every freeze-thaw cycle degrades it further. The single best habit is to aliquot:

  • Divide the reconstituted stock into single-use portions immediately.
  • Freeze the aliquots you are not using right away.
  • Thaw only what a given experiment needs.

This turns one vulnerable stock into many stable, single-use portions and keeps your material comparable across a research program.

Minimizing freeze-thaw damage

Repeated freezing and thawing is one of the leading causes of peptide degradation. Each cycle is mechanical and chemical stress. If a protocol has you returning to the same tube again and again, redesign it around aliquots. Track how many cycles any given aliquot has seen.

Document as you go

Record reconstitution date, solvent, concentration, and storage conditions alongside the batch number and its COA. When results look off later, this record is what lets you separate a material problem from an experimental one.
